Where is cleaved caspase-3?
cytoplasm
Caspase-3 exists in the form of pro-enzyme, and is located predominantly in the cytoplasm of cells.
Why cleaved caspase-3?
Cleaved caspase-3 propagates an apoptotic signal through enzymatic activity on downstream targets, including poly ADP ribose polymerase (PARP) and other substrates (2). In cell biology, caspase-3 antibodies that detect both uncleaved and cleaved versions of the enzyme are strong indicators of cell death induction.
Is caspase-9 cleaved?
The proform of caspase-9 has a low but measurable cleavage activity, but, when associated with the apoptosome, this is stimulated 2000-fold. Significantly, a non-cleavable caspase-9 mutant is also activated in an apoptosome-dependent manner with no loss of activity [5–7].
How do I activate caspase-3?
Caspase-3 is a cysteine–aspartic acid protease that cleaves cellular targets and executes cell death. Our current understanding is caspase-3 is activated by the cleavage of the interdomain linker and then subsequent cleavage of the N-terminal prodomain.
What is cleaved caspase 9?
Caspase-9 (ICE-LAP6, Mch6) is an important member of the cysteine aspartic acid protease (caspase) family (1,2). Cleaved caspase-9 further processes other caspase members, including caspase-3 and caspase-7, to initiate a caspase cascade, which leads to apoptosis (7-10). Duan, H. et al. (1996) J.
What is a cleaved caspase?
The cleaved-Caspase-3 (Asp175) antibody (referred to as cleaved-Caspase-3 antibody) from Cell Signaling Technology (Danvas, MA, USA) is a polyclonal antibody obtained from rabbit that was raised against a peptide in the large subunit of the human effector caspase, Caspase-3, amino-terminal to Asp175.
Why is caspase so named?
They are named caspases due to their specific cysteine protease activity – a cysteine in its active site nucleophilically attacks and cleaves a target protein only after an aspartic acid residue. Caspases also have a role in inflammation, whereby it directly processes pro-inflammatory cytokines such as pro-IL1β.
